Identification, mapping and relative quantitation of SARS-CoV-2 Spike glycopeptides by Mass-Retention Time Fingerprinting

Abstract We describe an analytical method for the identification, mapping and relative quantitation of glycopeptides from SARS-CoV-2 Spike protein. The may be executed using a LC-TOF mass spectrometer, requires no specialized knowledge glycan analysis exploits differential resolving power reverse phase HPLC. While this separation technique resolves peptides with high efficiency, glycans are resolved poorly, if at all. Consequently, consisting same peptide bearing different structures will all possess very similar retention times co-elute. Rather than disadvantage, we show that shared time can used to map multiple species location. In combination MSMS pseudo MS3, have constructed detailed mass-retention database glycopeptides. This allows any accurate LC-MS laboratory reliably identify quantify single overnight elastase digest in less 90 minutes.
